Hi Dan,
You may wish to consider the open source tool Blender (Blender.org) which works well in Windows Mac and Linux. It would be able to auto locate 2D markers to the skin spattered movie clip that you propose to use, and auto track their movement from frame to frame quite well. You can export the 2D position coordinates of the markers to a csv spreadsheet for later strain calculations with a short Python script.

Skin strain
Hi All,
I am starting to look in to measurement of skin strain. Initial research has indicated 2D digital image correlation with speckle tracking is likely the best approach, based on published data and available resources. There seems to be a range of free/open source software to facilitate the process, for example:
